Integrated course unit module: IN VIVO AND IN VITRO TESTING MODELS

Learning objectives

This core module aims to introduce students to the concepts of ex vivo ed in vitro cell models used to study pathological processes involved in the major diseases.

-Cell cloning
-Culture of animal cells

-Cell vitality and cell proliferation assays

Strategy of cell cultures
Biology of cultured cells.
Advantages and disadvantages of cell culture.
The environment of the cell culture: substrates, media, temperature, growth factors, etc.. "Scaling-up of cell cultures.
Kinetics of growth. Survival and toxicity.
Tissue dissociation and primary cultures.
Separation and characterization of specific cell types.
Maintenance of cultures.
Cryopreservation
Contamination of cell cultures.
The cancer cell: morphological and metabolic features.
Established cell lines.
Methods for transferring therapeutic genes
• transfection by physical methods: microinjection, electroporation, bombardment (gene gun)
• Lipofezione: liposomes, cationic lipid-DNA complexes
• viral vectors: retrovirus, adenovirus, adeno-associated virus, human herpesvirus.
