Learning objectives
Provide general principles of medicinal chemistry, evidencing the relationships between drug structure and their therapeutically relevant properties. Analyse the classification, the chemical, physicochemical and biological properties, the mechanism of action, structure-activity relationships and synthesis of main atiinfective and antitumor drugs.
Prerequisites
The exams of Organic Chemistry II and General Physiology must be passed before the examination.
Course unit content
In the first part, general themes will be addressed, including drug nomenclature, principles of pharmacokinetics, metabolism and mechanism of action, as well as basics on structure-activity relationships. The second part will be focused on the primcipal classes of antibacterials, antiprotozoan, antifungal, antihelmintic, antiviral and antitumor drugs.

1 - Parte generale
1.1 - Principi di chimica farmaceutica. Fasi dell'azione di un farmaco: fasi farmaceutica, farmacocinetica, farmacodinamica. Distribuzione dei farmaci. Reazioni metaboliche subite dai farmaci. Profarmaci. Meccanismo d'azione dei farmaci: teorie sull'azione dei farmaci; i recettori; enzimi ed inibizione enzimatica; altri bersagli molecolari. Proprietà chimiche e chimico-fisiche rilevanti per l'azione dei farmaci. 1.2 - Struttura dei farmaci ed attività biologica. Meccanismi d'azione specifici e non specifici. Isomeria ottica o geometrica ed attività biologica; isomeria conformazionale ed attività biologica. Isosteria e bioisosteria; gruppi aptofori e farmacofori. Relazioni quantitative struttura-attività.
2 - Parte sistematica. Antibiotici: betalattami; macrolidi; aminoglicosidi; tetracicline; ansamicine; polipeptidi; altri antibiotici. Antibatterici sistemici di sintesi: sulfamidici, nitroeterocicli, chinolonici, ossazolidinoni. Antimicobatterici. Antifungini: per uso topico e sistemico; antibiotici polienici, griseofulvina, tiocarbamati, allilamine, fluorocitosina, azoli, caspofungine. Antiprotozoari. Antielmintici. Antivirali. Antitumorali: agenti alchilanti; complessanti del DNA; antimetaboliti; antimitotici; inibitori delle topoisomerasi; inibitori di tirosin-chinasi; altri antitumorali.

Assessment methods and criteria
The student achievements will be evaluated by oral examination. Questions from the eaxminator and discussion will aim at establishing if the objectives of knowledge and comprehension have been achieved and if the studen is able to explain clearly and correctly the subjects. Through specific questions, the ability to apply student's knowledge to problems regarding medicinal, farmaceutical or toxicological chemistry for drug classes will be assessed.
